Jesper Klein nåede i løbet af sine 66 år at spille med i næsten 30 film, 14 tv-serier, herunder 9 julekalendere, og lagde stemme til 14 tegnefilm. Jesper Klein kender mange nok især fra TV 2's julekalender-klassiker, Pyrus. Her spillede han Bertramsen, der sammen med sin datter var forvaltere af rigsarkivet. Jesper Klein har også en søn ved navn Sebastian Klein, der ligeledes er skuespiller.
Jesper Klein døde efter at have været indlagt på hospice i en måned med en kræftsygdom. Han døde som sagt d. 22. august 2011.
